【預期用途】

尼古丁(Nicotine)是煙草中的主要生物堿，是導致吸煙成癮的物質動因，也是評價人體攝入煙草煙霧的常用指標。但因為尼古丁半衰期短，無法作為標志物檢測，其代謝物可替寧因為半衰期長作為吸煙和戒煙的標志物。

本品采用競爭抑制法和膠體金免疫層析技術，用于快速定性檢測人體唾液中的可替寧，適用于評價煙草煙霧攝入的初步篩查。

【主要組成成份】

• 可替寧檢測試劑盒（40人份）：每人份鋁箔袋單獨包裝。其中試劑盒由金標可替寧單克隆抗體、可替寧-BSA結合物、羊抗兔IgG多克隆抗體、硝酸纖維素膜、聚酯纖維素膜、塑料背襯、塑料模板組成。
• 一次性滅菌唾液取樣棒（40人份）
• 唾液收集器（40人份）
• 使用說明書（1份）

• 【檢驗方法】

• 撕開鋁箔袋，取出試劑，應在1小時內盡快使用。
• 拿出一次性唾液收集器。（步驟1）
• 手持收集器蓋帽使海綿頭含入口中，直至海綿頭吸滿液體（膨脹變軟）。（步驟2）
• 將海綿頭放回收集器并擰緊蓋帽，使所含樣本流入收集器下端。（步驟3）
• 將試劑盒置于干凈平坦的臺面上，同時打開滴樣孔蓋帽。（步驟4）
• 擠壓收集器上端管壁，垂直滴加2滴無空氣泡的唾液（約100ul）于加樣孔（S）中。(步驟5)
• 等待紫紅色條帶的出現，3-5分鐘時直接觀察結果，10分鐘后判定無效。

為了提高檢出結果，樣品可采取消化濃縮法處理，處理方法見附錄A(標準的附錄)。
3.1.1 痰
對牛常可用痰進行細菌學檢驗。牛咯痰極少，它在清晨采集。用硬橡膠管自口腔伸入至氣管內。外端連接注射器吸取痰液。亦可取牛咳出的痰塊進行檢驗。
痰樣品稀薄時，可加入等量的4%～6%硫酸(見附錄A中A1)處理，如痰樣品粘稠時，則加入5倍量的4%硫酸處理。充分搖勻后置37℃作用20min，以3000r/min～4000r/min離心，沉淀物作染色鏡檢、培養和動物試驗。
也可用2倍量15%～20%安替福民(antiformin)溶液(見附錄A中A3)處理痰液，將混合物置37℃作用1-2h后離心。以滅苗生理鹽水沖洗沉淀物幾次后，取沉淀物染色鏡檢、培養和動物試驗。
3.1.2 乳
懷疑有乳房結核時，可無菌采集乳汁進行檢驗(一般以擠出之zui后乳汁含菌量較多，早晨擠出的乳中含菌量zui高)。
將乳汁分置4-6支離心管中，每管10mL，以3000-4000r/min離心20-30min，吸取上層乳脂和管底的沉淀物作染色鏡檢、培養和動物試驗。
乳汁10mL加酒精和細菌各10mL及15％～20％安替福民溶液30mL，搖勻置37℃恒溫箱內1h～2h，取出加等量滅菌蒸餾水混勻，離心沉淀后棄去上清，取沉淀物作染色鏡檢、培養和動物試驗。
3.1.3 細菌和子宮分泌物
有細菌道結核可疑時，可采集公畜細菌、母畜子宮分泌物進行細菌學檢查。
處理子宮分泌物，可用15%～20%安替福民溶液，混勻后靜置2h～3h，3000r/min～4000r/min離心20min，棄上清，加10mL蒸餾水于沉淀物中。即可作動物試驗，經1.5～2個月剖檢。
3.1.4 尿
有腎結核可疑時，可采集尿液，一般采集中段尿液，以早晨*次尿為宜。
如僅作染色鏡檢，可將尿液以3000r/min~4000r/min離心20rain后，取沉淀物涂片。
如作培養或動物試驗，則應將尿液進行消化濃縮處理，再進行檢驗以免雜菌生長而影響檢驗結果。

3.1.1 sputum

3.1.2 milk

3.1.4 urine

For culture or animal experiments, the urine should be digested and concentrated, and then tested to prevent the growth of miscellaneous bacteria affect the test results.
