【預(yù)期用途】

尼古丁(Nicotine)是煙草中的主要生物堿，是導(dǎo)致吸煙成癮的物質(zhì)動因，也是評價人體攝入煙草煙霧的常用指標(biāo)。但因?yàn)槟峁哦“胨テ诙蹋瑹o法作為標(biāo)志物檢測，其代謝物可替寧因?yàn)榘胨テ陂L作為吸煙和戒煙的標(biāo)志物。

本品采用競爭抑制法和膠體金免疫層析技術(shù)，用于快速定性檢測人體唾液中的可替寧，適用于評價煙草煙霧攝入的初步篩查。

【主要組成成份】

• 可替寧檢測試劑盒（40人份）：每人份鋁箔袋單獨(dú)包裝。其中試劑盒由金標(biāo)可替寧單克隆抗體、可替寧-BSA結(jié)合物、羊抗兔IgG多克隆抗體、硝酸纖維素膜、聚酯纖維素膜、塑料背襯、塑料模板組成。
• 一次性滅菌唾液取樣棒（40人份）
• 唾液收集器（40人份）
• 使用說明書（1份）

• 【檢驗(yàn)方法】

• 撕開鋁箔袋，取出試劑，應(yīng)在1小時內(nèi)盡快使用。
• 拿出一次性唾液收集器。（步驟1）
• 手持收集器蓋帽使海綿頭含入口中，直至海綿頭吸滿液體（膨脹變軟）。（步驟2）
• 將海綿頭放回收集器并擰緊蓋帽，使所含樣本流入收集器下端。（步驟3）
• 將試劑盒置于干凈平坦的臺面上，同時打開滴樣孔蓋帽。（步驟4）
• 擠壓收集器上端管壁，垂直滴加2滴無空氣泡的唾液（約100ul）于加樣孔（S）中。(步驟5)
• 等待紫紅色條帶的出現(xiàn)，3-5分鐘時直接觀察結(jié)果，10分鐘后判定無效。

結(jié)核菌素皮膚反應(yīng)是遲發(fā)型細(xì)胞超敏反應(yīng)。它是抗原（結(jié)核桿菌或卡介苗）進(jìn)入機(jī)體使機(jī)體的免疫T淋巴細(xì)胞致敏，并大量分化增殖。當(dāng)已致敏的機(jī)體再次遭受到抗原入侵時，致敏淋巴細(xì)胞就會與之結(jié)合，引起細(xì)菌反應(yīng)性炎癥。表現(xiàn)在結(jié)核菌素注射部位形成硬結(jié)甚至發(fā)生水泡、壞死。結(jié)素試驗(yàn)陽性表明機(jī)體曾經(jīng)受到結(jié)核菌感染或接種過卡介苗。
一、牛型結(jié)核分枝桿菌PPD皮內(nèi)細(xì)菌反應(yīng)試驗(yàn)
用結(jié)核分枝桿菌PPD進(jìn)行的皮內(nèi)細(xì)菌反應(yīng)試驗(yàn)對檢查活畜結(jié)核病是很有用的。該試驗(yàn)用牛型結(jié)核分枝桿菌PPD進(jìn)行。
1、牛的牛型結(jié)核分枝桿菌PPD皮內(nèi)細(xì)菌反應(yīng)試驗(yàn)
出生后20d的牛即可用本試驗(yàn)進(jìn)行檢疫。
（1）操作方法
a) 注射部位及術(shù)前處理：將牛只編號后在頸側(cè)中部上三分之一處剪毛(或提前一天剃毛)，三個月以內(nèi)的犢牛，也可在肩腫部進(jìn)行，直徑約10cm。用卡尺測量術(shù)部中央皮皺厚度，作好記錄。注意，術(shù)部應(yīng)無明顯的病變。
b) 注射劑量：不論大小牛只，一律皮內(nèi)注射1mL(含2000IU)。即將牛型結(jié)核分枝桿菌PPD稀釋成每毫升含2萬IU后，皮內(nèi)注射0.1mL。凍干PPD稀釋后當(dāng)天用完。
c) 注射方法：先以75％酒精消毒術(shù)部，然后皮內(nèi)注射定量的牛型結(jié)核分枝桿菌PPD，注射后局部應(yīng)出現(xiàn)小皰，如對注射有疑問時，應(yīng)另選15cm以外的部位或?qū)?cè)重作。
d) 注射次數(shù)和觀察反應(yīng)：皮內(nèi)注射后經(jīng)72h判定，仔細(xì)觀察局部有無熱痛、腫脹等炎性反應(yīng)，并以卡尺測量皮皺厚度，作好詳細(xì)記錄。對疑似反應(yīng)牛應(yīng)立即在另一側(cè)以同一批PPD同一劑量進(jìn)行第二回皮內(nèi)注射，再經(jīng)72h觀察反應(yīng)結(jié)果。
對陰性牛和疑似反應(yīng)牛，于注射后96h和120h再分別觀察一次，以防個別牛出現(xiàn)較晚的遲發(fā)型細(xì)菌反應(yīng)。
